Shimadzu Nexera X2 UHPLC system
The Nexera X2 UHPLC system from Shimadzu showcases innovative technologies for improving UHPLC method development and operation. Solvent/buffer blending is achieved by utilising two quaternary gradient UHPLC pumps as the pre-mixers for a binary gradient delivery of the optimised phases. This allows optimising of pH, organic phase blends, ion-pair reagent addition and more.
When investigating analytical conditions during HPLC method development, selection of the mobile phase greatly influences the separation. In particular, when the analyte is an ionic compound, it is especially important that the mobile phase contains a buffer solution of appropriate pH because the analyte retention varies greatly depending on its ionic dissociation equilibrium. From an analyst’s perspective, however, the preparation of several mobile phases at various pH values for each analysis in the investigation of the analytical conditions represents a tedious and time-consuming effort.
Examples of such applications include the use of solvent blending to realise an optimised pH through phosphate buffer blending; and an ion-pairing application using TFA, where the TFA concentration is optimised by blending to give the best retention of angiotensin.
