The Olympus FluoView FV1000MPE Multiphoton laser scanning microscope system offers bright clear imaging in living cells at depth within specimens.

At the core of the FV1000MPE technology is the optical design. A custom AOM has been incorporated to adjust laser light intensity, with rapid on/off switching. This is combined with an automatic beam expander that ensures optimal adjustment of the excitation laser beam diameter and maximal filling of the entry pupil of the microscope objective when changing excitation wavelengths and objectives.

Correcting for light refraction in the sample for deeper imaging is essential. Olympus has designed a multiphoton-dedicated 25X objective (the XLPLN25XWMP) with a correction collar that can be adjusted to compensate for any refractive index mismatch, allowing the formation of an focal spot deep within the sample without reducing energy density.

The widefield design into both the FV1000MPE non-descanned detectors and XLPLN25XWMP objective captures the maximum amount of fluorescence signal for highly efficient imaging. A transmitted fluorescence detector with a dedicated NA condenser allows for bright fluorescence imaging deep within a live specimen and also allows concurrent second harmonic generation imaging.
