DIGESTIF is a generic standard designed to release indicators peptides which reflect the efficiency of digestion step on any bottom-up proteomics experiment. The product reflects the digestion properties of a complex protein mixture and is suitable for standardising a broad spectrum of applications, including LC-MS based proteomics, development of peptide mapping protocols, protein biomarker studies and controlling different proteases sources.

The product consists of a soluble recombinant protein scaffold, to which a set of 11 artificial peptides (iRT) of differing hydrophobicity with good ionisation have been incorporated. In the protein scaffold, the amino acids flanking iRT peptide cleavage sites were selected to either favour or hinder protease cleavage. After sample processing, the retention time and relative intensity pattern of the released iRT peptides can be used to assess the quality of sample workup, the extent of digestion, the performance of the LC-MS system and inter/intra-lab variability.
