The FormaPure system can be used to extract both total nucleic acid and total RNA and offers good yield and purity in an automation-friendly process. The technology can purify nucleic acids from a wide variety of tissue types for utilisation in techniques such as SNP genotyping, gene expression and sequencing.
By using the patented solid phase reversible immobilisation (SPRI) paramagnetic bead-based technology, the system does not require vacuum filtration, centrifugation or organic solvents such as phenol or xylene. Researchers can go from paraffin digestion to total nucleic acid extraction in about 4 h (without set-up) for 96 samples in a multi-well format using the company’s Biomek NXP or FXP Span-8 workstation. Recoveries are claimed to be consistently higher than competitive techniques.
The system can be used for total RNA or total nucleic acid isolation from FFPE tissue samples, and qRT-PCR† and microarray downstream application techniques.

The fixation of tissue samples in formaldehyde can cause extensive crosslinking of tissue components. Quality of nucleic acid will vary depending on a variety of factors, including how the formalin-fixation process was carried out, age of sample, storage conditions, etc.
