Fibrinogen-like protein 1 (FGL1), a molecule secreted by the liver and pancreas, is a major ligand of lymphocyte activation gene 3 (LAG-3). Recently, FGL1 has also been shown to be upregulated in human cancers. FGL1 binding to LAG-3 inhibits T cell response, where the FGL1-LAG-3 interaction blockade promotes tumour immunity by stimulating T cell expansion and activation. High plasma FGL1 levels are associated with poor clinical outcome in patients treated with anti-PD1 therapy.
