Regulation and activation of T lymphocytes depend on signalling by the T cell receptor (TCR) and also by co-signalling receptors that deliver negative or positive signals. The PD-1/PD-L1 signalling pathway is a negative regulatory mechanism that inhibits T cell proliferation and cytokine production. Blockade of the PD-1/PD-L1 interaction enhances anti-tumour immunity. The PD-1 pathway plays a major role in the inhibition of self-reactive T-cells.
