DNA Purification System

TOP (Trityl-on Oligonucleotide Purification), is a simple and high-throughput purification solution which combines a 96-well plate, a streamlined protocol, and a proprietary resin that will deliver optimum yield and purity for oligos up to 200 nmole in scale and up to 50 mers in length.

Oligonucleotides are the basic building blocks of genomic and proteomics research. Oligo purity is a concern for applications including single nucleotide polymorphism (SNP) detection or discovery, cloning, gel shift assays, ligation reactions, in-situ hybridisation, microarray fabrication, and longer oligos used in polymerase chain reaction (PCR) and sequencing.

After synthesis, a laboratory must purify the oligonucleotides by removing the reaction matrix and DNA sequences that failed during the synthesis process. The TOP product provides 90-95% pure oligonucleotides while at the same time delivering higher yields than many comparable purification techniques.

The TOP solution offers a combination of purification throughput and flexibility. TOP can purify up to 96 samples in 45 minutes and is fully compatible with liquid handling/robotic systems to provide a completely unattended, ultra-high throughput option.

The modular design of TOP makes it a 96-well plate product that matches the output of all currently available DNA synthesisers. The 96-well plate format easily adjusts to specific usage requirements by purifying anywhere from one to 96 samples simultaneously. And, the modularity of the Varian TOP product reduces purification costs by eliminating wasted wells. TOP relies on an easy-to-use gravity flow process that results in more reproducible results and lowers overall costs because no vacuum manifolds are needed. The simple eight-step procedure uses one-third the solvent volume of other products and requires only a single pass of the sample, offering improved compatibility with robotic systems for high throughput laboratories.
