Tomocube HT-X1 Plus holotomography and spinning disk confocal microscopy system

The HT-X1 Plus holotomography (HT) system from Tomocube has been integrated with the X-Light V2 spinning disk confocal microscope (SDCM) from CrestOptics to offer a spinning-disk confocal holotomographic imaging system. The HT-X1 Plus has been named one of the top 10 microscopy innovations in the 2025 Innovation Awards by Microscopy Today.

This system combines the power of holotomography with fluorescence to enable precise molecular-structural correlation in live-cell and whole-organism experiments, unlocking the full potential of fast, gentle 3D correlative imaging, and is available from Tomocube.

Holotomographic imaging involves illumination of live cells with low-power visible light from multiple illumination angles, followed by measurement of the phase delay of the transmitted light. The technique uses the measured refractive index as imaging contrast, allowing visualisation of 3D living cells and tissues without labelling or staining. With its high spatial resolution capable of capturing subcellular organelles, users have increasingly sought confocal fluorescence capabilities to design multiplex imaging workflows that minimise photodamage.

By combining two imaging modalities, researchers are able to visualise cellular structures in three dimensions with high clarity. By providing quantitative phase information, holotomography captures structural details that would otherwise be difficult to observe, offering a deeper understanding of cellular morphology and dynamics. Additionally, the integrated platform is designed to provide precision alignment of holotomography and fluorescence-based imaging datasets, allowing researchers to easily study molecular and phenotypic data in tandem and generate more powerful insights across biophysical imaging applications.
