Understanding how biomolecules interact is fundamental to drug discovery, biologics development and biochemical research — but weak, transient or low-abundance interactions can be difficult to characterise using conventional techniques.

The Creoptix WAVEsystem from Malvern Panalytical provides highly sensitive, real-time, label-free analysis of biomolecular interactions using patented grating-coupled interferometry (GCI). Unlike traditional approaches that rely on fluorescent or enzymatic labels, the WAVEsystem directly measures interactions at the sensor surface, delivering kinetic rate constants (ka and kd), affinity (K_D) and binding responses.

The system’s no-clog WAVEchip microfluidics can enable analysis of complex or crude samples, reducing the need for extensive sample purification. Its high sensitivity and high signal-to-noise make the system particularly valuable for challenging applications, including small-molecule and fragment screening, weak binders, antibody characterisation, protein–protein interactions and interactions involving large ligand-to-analyte molecular weight differences. WaveRAPID can accelerate kinetic screening by determining interaction kinetics from a single sample concentration, reducing sample consumption.

The new KYMAchip has been designed to deliver fast signal transitions, faster kinetics and greater reproducibility. Its optimised channel design enables measurement of rapid association and dissociation events, while reducing transition artefacts and channel-to-channel variability. Like the WAVEchip, KYMAchip supports challenging samples while offering the flexibility to remove, store and reuse the chip — helping researchers get kinetic data from every experiment.
