Random labelled single-stranded RNA probes can be synthesised by in vitro transcription. The reaction is catalysed by bacteriophage T7 RNA polymerase that incorporates labelled NTPs (mostly UTP and CTP) as substitute for their natural counterpart using linear, RNA probe-encoding DNA as template.

Jena Bioscience’s HighYield T7 RNA Labeling Kits can achieve individual optimisation of labelled NTP/NTP ratios in a single nucleotide format. The optimal ratio of labelled NTP/NTP in terms of product yield and labelling efficiency depends on both the type of labelled NTP and final application.

The kits are designed to offer maximum optimisation flexibility in contrast to fixed NTP labelling mixes, combined with high product yields. The combination of (poly)-HRP-conjugated biotin/digoxigenin detection reagents with labelled tyramides further increases detection sensitivity of up to 100-fold.

The products are available in the following labels for UTP-based kits: FAM, Cy3, Cy5, AF405, Atto 488, AF488, AF555, STAR 580, AF594. AF647, STAR RED, IR680LT, IR750, Digoxigenin, Biotin and Azide. CTP-based kits are also available for AF488, AF555, AF594 and AF647 labels.
