IDT mutagenesis application guide

Integrated DNA Technologies (IDT) has released its free to download mutagenesis applications guide. Providing an overview of in vitro mutagenesis, along with in-depth protocols and troubleshooting advice, the guide makes it easy to carry out efficient and effective experiments.

Supplying a wealth of useful details, the guide describes traditional mutagenesis approaches, including site-directed mutagenesis by polymerase chain reaction (PCR), primer extension, inverse PCR and cassette-based methods. The guide also covers random mutagenesis, a technique which is often used to optimise protein expression or function by creating unpredictable alterations.

One approach to mutagenesis detailed within the guide is the use of IDT’s Ultramer oligos, which simplifies the addition of large changes into a target sequence, providing greater experimental flexibility. This is achieved since Ultramers are said to be the highest fidelity long nucleotides currently available, with lengths varying from 60-200 bases. As such, they enable primers to be extended into regions that have traditionally been difficult to target, with more optimal sequence composition than was previously attainable.
