The next generation of immunoassay reagents is being engineered through linking oligonucleotides to antibodies. Immuno-PCR combines the specificity of antibodies with the amplification power of PCR, allowing a 10- to 1000-fold increase in sensitivity. Antibody-oligonucleotide conjugates also have the potential to be a platform tool in multiplexed protein diagnostic assays.

Using standard chemistry and methodology, the linking of an oligonucleotide to an antibody is difficult, time-consuming and requires specialist knowledge. To overcome these issues, the Thunder-Link oligo conjugation system has been designed to allow the non-specialist to generate antibody oligonucleotide conjugates easily and efficiently.

All necessary components for a successful conjugation are supplied. The inclusion of positive controls enables the end user to confirm that the conjugation chemistry is working correctly.

The end user chooses the oligo sequence and antibody. Oligo sizes (HPLC purified) are between 10-120 bases. The technology is unidirectional and thus only allows the formation of antibody-oligonucleotide complexes - never antibody-antibody or oligonucleotide-oligonucleotide.
