Epoch multi-volume nucleic acid quantification application note

25 May, 2011 | Millennium Science Pty Ltd

Nucleic acids isolated from a variety of sample types such as animal models, body fluids or eukaryotic and bacterial cells provide the starting material for a broad range of downstream applications including gene expression, genotyping, sequencing and cloning. Quantification of isolated nucleic acid is necessary prior to introduction into these applications. The Epoch Spectrophotometer System provides a means of obtaining accurate measurement over a wide range of sample concentrations and volumes, including microvolume quantification. Comparison to other microvolume dedicated devices, such as NanoDrop, shows equivalent performance.

Millennium Science cAMP Alphascreen application note

25 May, 2011 | Millennium Science Pty Ltd

Adenosine 3, 5 cyclic monophosphate (cAMP) plays a critical role in the transmission of signals by functioning as a ‘second messenger’. Binding of hormones or ligands to their respective receptors can either inhibit or enhance the production of cAMP by changing the activity of the enzyme adenylate cyclase. Adenylate cyclase is a membrane-associated enzyme that catalyses the formation of cAMP from ATP. Intracellular cAMP levels, in turn, regulate enzymatic activity of numerous protein kinases, which phosphorylate specific targets setting off a cascade of cellular events. Because of the importance of cAMP as a cellular messenger, several different reading methodologies have been developed to measure cellular levels, one of which is AlphaScreen. Here we describe and demonstrate the ability of the Synergy 2 to quantitate cAMP using this methodology.

BioTek HTRF AKT application note

25 May, 2011 | Millennium Science Pty Ltd

AKT is a serine/threonine protein kinase that plays a key role in cellular processes. AKT has become a popular target for drug discovery campaigns, due to the fact that AKT inhibitors may help to treat a number of cancers. BioTek has demonstrated an automated homogeneous assay to probe AKT phosphorylation at its serine 473 residue using endogenous levels of kinase expression within human primary HUVEC cells.

BioTek cytokines ELx405 washer application note

25 May, 2011 | Millennium Science Pty Ltd

Cytokines and growth factors are cell signalling proteins that mediate a number of physiological responses. In addition, they are also associated with a number of diseases including tumour growth and metastasis, infections and inflammation. Because these molecules rarely act unilaterally it is important to assess a number of them in parallel. The advent of multiplex assays using Luminex xMAP bead technology has simplified the task of performing multiple assays on samples. BioTek has demonstrated how to automate the wash steps of the Bio-Rad HumanCytokine 8-plex assay kit using the ELx405 Magnetic Bead Washer.
